Pregnenolone Description and Effects

In this detailed description of Pregnenolone we learn that Pregnenolone is an important precursor hormone that has been shown to have beneficial effects on memory and in allieving auto-immune illnesses. Although studies began in the 1940s there has been relatively few studies done in recent years, so the working of Pregnenolone is not fully understood. It may interfere with the action of drugs prescribed for seizures or depression, and its effects on dependant hormones has not been well studied.

In a study of healthy men in their twenties, it was found that just one mg of Pregnenolone daily was enough to improve the quality of their sleep, and decreased intermittent wakefulness. This is the opposite effect from larger doses, which have been found to cause excitation and sleeplessness.

Another study found that 25 mg of Pregnenolone administered twice daily improved the performance of army pilots under stressful conditions. Pregnenolone has been found to be significantly lower among individuals with depression than among healthy people.

For the treatment of auto-immune diseases, this report says:

Researchers have reported on the use of pregnenolone in a variety of rheumatologic diseases. In a study of pregnenolone therapy (intramuscular injection, 50–600 mg daily) for rheumatoid arthritis, six of eleven individuals experienced moderate to marked improvement in symptoms of joint pain and joint mobility. The symptom improvement was apparent two to four days after therapy was initiated. In a study of thirteen adults with osteoarthritis, pregnenolone therapy reduced the pain and improved the range of motion in seven of the study participants. Pain recurred when therapy was discontinued. In a person who suffered from gouty arthritis and was unresponsive to traditional medications, pregnenolone therapy resulted in a dramatic response within three days of initiating therapy. ... Of the 59 individuals reported in this paper, the only adverse effect was redness or pain at the site of injection. No systemic adverse effects were reported.

There is no mention, however, of how long the studies were, so it is not clear if there may be long-term effects.

Many studies show a U shaped distribution of Pregnenolone effects, so that small doses provide benefits, larger doses have little effect, and even larger doses are again beneficial. This might be explained by the fact that there is a feedback-loop at play. Small doses stimulate a reaction, but larger doses are dampered by feedback effects, unless the dose is so large that it overwhelms the feedback mechanism.
